Folylpolyglutamate synthase, mitochondrial is an enzyme that in humans is encoded by the FPGS gene.

This gene encodes the folylpolyglutamate synthetase enzyme. This enzyme plays a major role in creating and maintaining as cytosolic and mitochondrial folylpolyglutamate concentrations and, therefore, is essential for the homeostasis of folic acid and survival of proliferating cells. This enzyme catalyzes ATP-dependent addition of glutamate fragments to folic acid and its derivatives folic acid. While several variants of the transcript may exist for this gene, the full-length nature of only two have been biologically tested to date. These two variants encode different isoforms.
